Effect of Ultraviolet Irradiation and Heating of Phage on Phage Hemagglutination


Abstract
The author studied on the hemagglutination-forming ability of phage which is heavily irradiated by ultraviolet ray and heated at various temperatures. For this experiment, the Klebsiella phage-host cell system and ultraviolet ray of 2537A were used. The following results were obtained.
The hemagglutination-forming ability of phage was destroyed by heavy irradiation of ultraviolet ray and, the more the ultraviolet was irradiated, the more remarkable became the degree.
The hemagglutination-disappearing velocity of sensitized red cells, to which the ultraviolet irradiated phages were already adsorbed, by keeping at warm temperature was a little slower than that to which normal phage was adsorbed.
The normal phage could be adsorbed to the sensitized red cells pretreated with ultraviolet irradiated phages against that pretreated with normal phage and cold phage hemagglutination by the former red cells was a little stronger than that by the after one.
The cold hemagglutination-forming ability of phage heated at 60¡ÆC for 20 minutes was similar to that of normal phage had analogous surviving titer. However, the hemagglutinationform_ing ability of phage heated at 65¡ÆC for 20 minutes was considerably weaker than that of normal phage had similar surviving titer.
When phage suspension is heated at various temperatures, respectively, if the surviving titer of each phage is similar, the hemagglutination-forming ability of each heated phage was also similar regardless of heating conditions.
The normal phage could be adsorbed to the sensitized red cells pretreated with heated phage and the h emagglutination-disappearing velocity of sensitized red cells, to which heated phage was already adsorbed, by keeping at warm temperature was similar to that to which normal phage was adsorbed.
